摘要:
为探索细胞力学响应能力,本研究利用荧光共振能量转移(fluorescence resonance energy transfer,FRET)技术继续检测细胞生物力学通讯的信号变化。结果显示,气道平滑肌(airway smooth muscle,ASM)细胞在基质胶(Matrigel含0.5 mg/mL I型胶原)上比在玻璃基底上表现出显著增强的运动性能,并促进群体分枝形态发生。通过FRET技术检测发现,与玻璃上的对照细胞组比较,基质胶上的ASM细胞中ERK和Cdc42活性出现了显著下降,代表胞内收缩信号的RhoA活性有一定下调。这些结果提示在ASM细胞力学通讯和分枝形态形成中,与细胞增殖反应相关的ERK和Cdc42信号受到一定的抑制。
Abstract:
We used fluorescence resonance energy transfer (FRET) to study cell signals during the mechanical communication. Airway smooth muscle (ASM) cells showed apparently enhanced mobility on the hydrogel (Matrigel containing 0.5 mg/mL type I collagen) than on the glass, leaded to branch assembly. Monitored by FRET biosensors, ERK and Cdc42 signals had significant decreases in ASM cells during culture on the hydrogel in comparison to the glass, while RhoA activity had modest decrease. These results implicate that cell proliferation-related ERK and Cdc42 signals are attenuated during the mechanical interactions and branching assembly in ASM cells on the hydrogel.
